Scientists from Georgia Tech and Vanderbilt University have developed a miniature “bone marrow-on-a-chip” that mimics key features of human bone marrow. The model allows researchers to observe how antibody-producing cells move, settle and survive within different bone marrow environments.
The system combines a human lymphoid organoid with a vascularised microfluidic chip containing two important bone marrow niches. Researchers observed antibody-secreting cells migrating through the model and accumulating around blood-vessel-like regions, with some showing a “stop-and-go” movement linked to the CXCR4–CXCL12 signalling pathway.
Published in Science Advances on September 11, the study could help researchers better understand how long-lived plasma cells maintain antibody production after infection or vaccination. The model may also support research into ageing, allergies, autoimmune diseases and certain blood cancers, although it remains a laboratory research tool and does not fully replicate human bone marrow.
